Effect of Tamoxifen

Some cancers are promoted in their formation and growth by hormones. This is, for example, the case of breast cancer, whose development and spread is dependent on the female sex hormone estrogen. The estrogen attaches to special hormone receptors on the cancer cells and stimulates them to grow.

Tamoxifen belongs to the drug class of antiestrogens and typically blocks the binding of estrogen to the hormone receptor of cancer cells. Due to the lack of growth signal by the hormone there is a decrease in cell proliferation. In the case of metastatic breast cancer, treatment with tamoxifen causes complete or partial regression in about 30% of cases, especially of the secondary tumors in the soft tissues and bones. The probability of recurrence is even around 50 to 60 percent if the presence of estrogen receptors in the tumor tissue has been proven.

In early-stage breast cancer, prolonged non-relapse and survival time can be achieved by tamoxifen therapy after surgery.

Tamoxifen itself does not inhibit the growth of cancer cells. The active substance is converted into the active form endoxifen after absorption into the body in the liver by the enzyme CYP2D6.

    After almost a year, severe side effects now occur. Am after a year Nolvadex Zoladex – stiff in the joints and back. Have strong back pain, especially when lying or bending over. My tendons hurt and shorten, especially the Achilles tendon hurts strong. I can not walk as I did a year ago. Furthermore, I have ongoing eye infections and I am steadily gaining weight although I am already very careful.

    What worries me are the hot flashes. I sweat so much that one might think I came out of the shower !! I also have sleep problems or can fall asleep very badly. But I can live with that. If this reduces the risk of getting breast cancer again, I am happy to accept this because otherwise I am doing very well, whatever I wish you to do.

    I am in my mid 30s, the side effects increase from year to year: hot flashes at least 20 times a day, circulatory problems, depression, insomnia, tiredness, lack of motivation, lack of concentration, hair loss, …. But thank goodness, nothing has increased. I will still take the medication to avoid relapse.
