Applications of the active substance trastuzumab

Trastuzumab is a cancer treatment for breast cancer and is used differently in different degrees of disease.

In the early stages of cancer, trastuzumab is used alone after surgery, radiotherapy or initial chemotherapy. However, the prerequisite is that the binding site HER2 can be detected in the laboratory on the surface of the breast cancer cells. Trastuzumab needs to dock with these in order to take effect. Following the evaluation of new studies, the use of trastuzumab is now also approved for chemotherapy in patients with early detected HER2-positive breast cancer.

Another field of application for trastuzumab is breast cancer, which has already formed secondary tumors (metastases). As an individual therapy, however, the drug is used only after a previous chemotherapy or at. This chemotherapy must have included at least two cytotoxic drugs (from the subgroups of anthracyclines and taxanes), unless this treatment was unsuitable for the patients. In addition, in patients with hormone-sensitive cancer, a previous hormone treatment must be unsuccessful or such a treatment was not suitable for the patient.

If patients have not yet received chemotherapy for their cancer, trastuzumab is used as a combination partner to cytostatic drugs such as paclitaxel and docetaxel.

In postmenopausal women who have not yet received trastuzumab and have a hormone-sensitive breast cancer, trastuzumab can also be used in combination with a so-called anti-estrogen or an aromatase inhibitor such as anastrozole.

Trastuzumab is also being used to treat cancers (adenocarcinomas) associated with gastric malignancies and the esophageal junction in patients who have not previously received cancer treatment for their disease. Again, the precondition is that the cancer cells have the HER2 binding site. Trastuzumab is used in combination with other cytotoxic drugs such as capecitabine or 5-fluorouracil and cisplatin.

Mode of action of trastuzumab

Trastuzumab is one of the largest group of immunological drugs. More specifically, trastuzumab is a recombinant humanized IgG1 monoclonal antibody against human epidermal growth factor 2 (HER2).

Antibodies are proteins that are actively involved in the body’s defense. Monoclonal antibodies are artificially produced and produced by a cell line derived from a single B lymphocyte (a special form of white blood cells). These monoclonal antibodies bind specifically to a single, unique protein structure on the surface of other cells. In the case of trastuzumab, this is the human epidermal growth factor 2 (HER2), which is produced by 20 to 30 percent of breast cancers and also some cancer cells at the stomach or esophagus junction and is characterized by a particularly rapid course of the disease.</p >

Both in the test tube and in animal experiments, trastuzumab has been shown to inhibit both the growth and proliferation of human cancer cells that produce HER2. In addition, trastuzumab highly effectively marks these cancer cells so that they can be better recognized and destroyed by the body’s own defense.

    strong dizziness since Herceptin. Since taking Tamoxifen and Herceptin I have a headache, shivering, weight loss, currently.     Every three weeks since December I have been receiving the drug Herceptin.     During my 2007 Herceptin Therapy, I gradually developed circulatory problems, high blood pressure, and also cardiac arrhythmias, which are still not resolved. I am currently getting ACE inhibitor 10mg. I would be interested to know if my complaints really come from the Herceptin. After my breast cancer surgery, chemotherapy and radiation, my body and soul have been through a lot. Maybe there are several factors together.

    Actually, I can not say much about the effectiveness, because I still get it now and you have to wait and see if there is no new tumor. But Herceptin gives a very good chance of getting a grip on this rather aggressive type of cancer and is also quite well tolerated compared to chemotherapy.     I got EC 4 times and Taxotere 4 times each with Herceptin, had some side effects, especially after taxotere, such as respiratory distress, bone pain, nails fragility. But the success speaks for itself, my tumor has completely receded and only the tumor bed had to be removed. Everything was worth it and it was also very bearable. Now I get to prevent a recurrence for another year of herceptin and new side effects I have until now no. I can only try to encourage any woman, because as bad as the chemotherapy is often described, it was really not for me, she was a fly-shit to what I was previously reported.     have just described my experience, comes to this: bone pain, muscle weakness, especially after Zometa (1/4 a year) for 2 nights chills and body aches, after taking ibuprofen better. Also gaining weight overall by taking tamoxifen and Herceptin (treatment ended on 13/03/2008)

    After 3 FEC and 3Doxetacel chemos at intervals of 3 weeks each I received 28 (+ 8 times Boost) irradiation, then 1 every 3 weeks of herceptin infusions, then 2 years Zoladex, and must now take 5 years of tamoxifen tablets.     Strong burning pain in both feet, sometimes I can barely walk. Weight gain, movement limitations of the fingers. Water retention in the hands and feet, but no impairment of the heart.     I have been receiving Herceptin as an infusion every 3 weeks since the beginning of the year. At first I had no problems – but since 6 weeks I have deaf hands u. Feet. I’m listless and run like drunk. In addition, I have pain in all bones.

    From July 2003 to July 2007, I received hercepetine 1 x weekly via the port, accompanying 8 chemos after mamma ca, left breast and 8 lymph node removal. 2005 recurrence, again 8 chemocycles and always in between and again accompanying hercepetine. Dizziness, numbness in toes and fingers, severe weight gain, soft tissue pain, listlessness, loss of weight in the sense of weight, inflammation in the legs. I then canceled the treatment, which was to last for life, after obtaining a second opinion. Subsequently severe bone and joint pain and unsuccessful arthroscopy with restricted walking. Weight has remained, but I feel much better overall.
