Action of Amitriptyline

Amitriptyline belongs to the active ingredient group of tricyclic antidepressants and also has its mode of action.

All brain nerve reactions are mediated by messengers such as serotonin and norepinephrine. These messenger substances are stored in small vesicles at the nerve endings and are released on excitement of the nerve. In this way they reach other nerves and bind to special binding sites (receptors). The affected nerves then transmit the stimulus signal. Once they have fulfilled their task, the messenger substances are taken back into their storage via special transport systems or degraded by enzymes.

Amitriptyline has a dual effect. Once the active ingredient in the brain occupies different binding sites for messenger substances and thus changes the influence of these messengers on the brain metabolism. Secondly, amitriptyline blocks transport substances that return the messengers serotonin and norepinephrine to their storage sites after signal transmission. As a result, serotonin and norepinephrine remain longer at the site of action and their effectiveness increases. Amitriptyline in this way triggers anxiety and tension states and reverses depressive moods. The soothing effect of amitriptyline usually occurs very quickly, while the mood-improving effect begins after about two weeks of regular use.

Chronic pain often interferes with the processing of pain signals in the brain. The messenger serotonin plays a major role in building a filter for incoming pain signals. By influencing serotonin, amitriptyline can replace the constant use of painkillers. The active ingredient must be taken for pain relief but then regularly over a long period of time.

    I had to take Saroten early in the evening so I could get out of bed early in the morning. If I did not sleep for 8-9 hours, I was groggy all day. If I had enough sleep, however, I was well rested and fit. Headaches are almost gone, tinnitus as well.     Elavil for Depression

    I was prescribed Saroten after taking citalopram for years. Due to various causes (for example, hardly any therapeutic measures) it was decided to prescribe Saroten (100 mg, retard) during a hospital stay. You get used to a lot of things and so I think that after several years of taking Citalopram it just lost its effect. Unlike Citalopram, I take Saroten in the evening instead of in the morning. At the clinic I got it at about 22 o’clock. About 2 hours later I became very sleepy and could also sleep well. I’ve been doing better since Saroten, but since the last hospitalization I have changed my life quite a bit to finally get rid of the depression. One should never forget that antidepressants are just an adjunctive support towards improving or curing a depressive phase. A regular daily routine and sports are prerequisites for improvement. A tip: Saroten has quite a sedative effect that can lead to the fact that you get out of bed in the morning, is a little dazed or does not even notice the alarm clock. If this happens to you, I recommend you to take it a little earlier. I take the tablet after some try now at 18 o’clock, so I am tired at 22 o’clock, sleep in time and get well out of bed in the morning. In any case, you should also discuss this with your doctor.

    For 12 years I was prescribed by the family doctor Saroten 50mg and Xanax 0.25mg for depresions and sleep disorders. Now I have been suffering from anxiety disorders for six weeks, my family doctor then referred me to a psychiatrist. Now I take 50mg Trittico in the evening and 20mg Cipralex in the morning.     I have had experiences with three medicines, which I took one after the other. First of all citalopram, saroten (amytriptilin) and tianeurax (tianeptin) belong in this order. Reason for my treatment is a 13 months continuous dizziness that is still my daily companion and is the cause of my depression. I try to write as much as possible about all medications. These are different for each of me it was like this. 1. Citalopram. Taken for 3 weeks. A new generation serotonin reuptake inhibitor with a drive-enhancing component. My doctor recommends one tablet in the evening before going to bed. Of this I can only advise against it is impossible to sleep after that the drive-increasing effect occurs very quickly. That’s why I always took those in the morning. The sleep rhythm is a bit confused. Already in the first days you can feel the drive increase significantly. the mood-enhancing effect did not really work for me but some side effects. To mention would be there, for example the inner restlessness, sleep disturbances, mood swings and motivational problems. The drive increase is not always advantageous. It often happened that I just had to walk around the apartment because I could not sit still. All in all quite bearable but only if the desired effect can be achieved. I think the drive increase did not have to be that when the mood improves that comes automatically. Citalopram is one of the antidepressants with very little side effects. However, there is an improved version called: escitalopram supposedly with even fewer side effects and better effects. 2. Saroten (amytritilin). 4 weeks ingestion. Tieneptin is the only known drug that enhances serotonin uptake. In fact, he does exactly the opposite of what the other antidepressants do. But the secret is probably somewhere else. Anyway, I am very pleasantly surprised. I take the drug now for 10 days daily 3 tablets (12.5mg). For me, the effect occurred after a week already. I’m in a very good mood, I’m motivated and doing the things that I used to put off, like sorting paperwork on my desk. I have fun keeping up my social contacts and can even do sports again without any side effects hindering me. I’m optimistic again and emotionally balanced. It is neither impulsive nor sedative and I appreciate that very much. The taste of food is somehow more intense. I feel that as positive. The first night I had a funny dream. Otherwise I have no side effects so far. That’s how I imagine the effects of an antidepressant. It looks very inconspicuous. Unfortunately, my dizziness is still unchanged, but now I have reason to hope for improvement. A little tip on the edge for those who have to deal with depression. Short-term relief and immediate mood-enhancing effects can be achieved with a completely sleepless night. I tried it myself.
