Action of Leflunomide

The active substance belongs to the group of antirheumatic basic therapeutics. This group is also referred to as a group of Disease Modifying Antirheumatic Drugs (DMARD). This English name translates as: disease-altering antirheumatic agents. Thus, these substances are delimited against active ingredients that only relieve the symptoms of the disease.

Joint rheumatism leads to increased formation of endogenous defense cells (T-lymphocytes) and T-cell-dependent autoantibodies (destroying the body’s own cells). These blood cells and antibodies destroy the joints. Leflunomide is a so-called prodrug, so a precursor substance. It is converted in the body to its actually active ingredient, the malononitrile A771726. Malonitrilamide inhibits the enzyme dihydroorotate dehydrogenase (DHODH) in humans. However, this enzyme is necessary for the proliferation of rapidly dividing cells, especially the lymphocytes important for the immune defense. An inhibition of the enzyme prevents the proliferation of lymphocytes and the associated joint destruction. Leflunomide is better tolerated by the stomach than its active ingredient.

On the one hand leflunomide has a stimulating effect on the body’s defense, on the other hand it has an immunosuppressive effect, that is, it suppresses the body’s own defense reactions. The latter can result in significant side effects.

    Achilles tendon ached suddenly and so severely that I could barely walk. Shoulder joints or tendons ached suddenly and so severely that I was severely restricted in my mobility. Diarrhea (several times a day), sometimes very severe itching (chest), loss of appetite. In addition, sleep disorders. After weaning, the diarrhea disappeared after a few days. The itching also stopped. Pain on the Achilles tendon and shoulder joints also improved daily (within 4 weeks). After renewed use (by doctor with dose reduction 20mg every 2 days) came after a few hours (about 12 hours) again the violent skin itching at chest level.

    After about 6 weeks strong, sometimes uncontrollable diarrhea – severe tiredness and loss of appetite. After three months blisters, redness and fistulas on the oral mucosa. Pain on the tendon attachments of some joints. Thereafter drug discontinued.

    Take Arava since 05.06.08 and I am very satisfied with it. After several years of pain and relapses finally almost no pain. The mobility is already very good in the morning. Almost no joint problems anymore. The knight armor is gone. Everything abschwollen. No more thick joints (especially the knees as with a Preisboxer). Can not believe it. My quality of life is almost 100% now. Swallow the small pill in the evening before going to bed, so the side effects have no chance. Have a few small red skin. Not worth talking about. I am very happy that after 10 years, it is finally back up.

    Arava for Arthritis

    I have rheumatism since I am 18 years old. I’ve been taking Arava since May 2007. Since then I’ve been doing very well, hardly able to move before. My rheumatologist first wanted to prescribe MTX for me, since I refused to take it she suggested to me Arava. I am very satisfied, have no side effects at all and my quality of life has increased by 100%.
